Aceto-Orcein Stain

|
Warm gently (do not boil), Used to stain chromosomes in chromosome squashes of Drosophila salivary glands. Stain for 2-5 min. prior to making the squash. |

Mayer’s Carmalum
|

|
|
| Boil 20 minutes. | ||
| Working solution: | ||
| carmalum stock solution |
5 ml
|
|
| glacial acetic acid |
0.4 ml
|
|
| distilled water |
100 ml
|

Directions |

|
Mix ingredients for working solution. When dissolved, filter. Add 1 ml formalin to inhibit mold growth. Will keep indefinitely. Stains nuclei. Is a progressive stain. Specimens are usually adequately stained in 4-8 hours. Stop staining when specimen is a deep pink-red color. |
